What is Grandaxin?

Grandaxin (trade name Tofisopam) is an anxiolytic drug used in Europe and Russia. He belongs to the family of benzodiazepines. However, grandaxin has a different pharmacological effect than other benzodiazepine tranquilizers.

The main active ingredient of Grandaxin is tofizopam, which is a crystalline powder of a yellowish hue.

It does not bind to benzodiazepine receptors, does not cause dependence and does not possess amnesic or muscle relaxant effects in the therapeutic dosage.

The reason for such differences is the chemical structure of Tofizopam. This is a derivative of 2,3-benzodiazepine, which differs from 1,4-benzodiazepines (for example, alprazolam, diazepam or phenazepam) at the position of nitrogen atoms.

Grandaxin is sold as an anxiolytic remedy since the 1970s. To date, it is still popular in Europe and CIS countries as an alternative to benzodiazepines. This drug is not approved for use in the United States, however its D-enantiomer Dextofisopam is in phase II clinical trials for the treatment of irritable bowel syndrome.

Indications for use

Official indications for the use of grandaxin are:

  • Neuroses and neurotic diseases (conditions accompanied by emotional stress, anxiety and apathy).
  • Reactive depression.
  • Violation of regulation and post-traumatic stress disorder.
  • Climacteric syndrome.
  • Premenstrual syndrome.
  • Alcohol withdrawal syndrome.
  • Myasthenia gravis, myopathy, atrophy of neurogenic muscles and other conditions with secondary neurotic symptoms, when treatment of muscle relaxant anxiolytics is inappropriate.

According to studies, this drug can also be used to treat dizziness, the appearance of tinnitus. Several clinical trials conducted in Japan indicate that Grandaxin is an effective treatment for these conditions.

Side effects and contraindications

Grandaxin is usually tolerated better than typical benzodiazepines. It is not addictive and does not contribute to the development of sedation. Nevertheless, the drug may cause adverse effects. Possible side effects provided by the manufacturer include:

  • Loss of appetite.
  • Constipation.
  • Flatulence.
  • Nausea.
  • Dryness in the mouth.
  • Headache.
